README These files contain the raw sequencing reads and mapped assemblies for the Nematostella vectensis BAC sequencing project, as described in the paper: Antje H.L. Fischer, Sarah Tulin, David Freedman, and Joel Smith. 2013 Employing BAC-reporter constructs in the sea anemone Nematostella vectensis. Integrative and Comparative Biology. The sequenced libraries contained DNA sequence from 5 Bacterial Artificial Chromosomes (BACs) engineered to contain ~200,000bp of genomic sequence from the Nematostella genome containing the genes Nv-Dpp, Nv-Foxa, Nv-Wnt1, Nv-Wnta, and Nv-Chordin. raw reads.zip :: This file contains the raw, untrimmed reads from two assembly runs as zipped qseq files. Created by sequencing 100bp single end reads on and Illumina HiSeq 1000. mapped_raw_reads.zip :: This folder contains two files created by mapping the trimmed read set against the Nematostella genome using the CLC Bio genomics workbench version 5.1 NGS tool mapper. There is a sam file and an indexed sam file. When placed in the same folder, they can be viewed as tracks on top of the Nematostella genome in a viewer such as the Broad Insitute's Integrative Genomics Viewer (http://www.broadinstitute.org/igv/). When unzipped, the sam file is quite large, ~30GB, and will take extra time to load in IGV. de_novo_assembly.fastq :: This file is the de novo assembly of the BAC sequences created using CLC Bio genomics workbench version 5.1 NGS de novo assembler. de_novo_assembly_mapped.zip :: This folder contains two files created by mapping the assembled contig set against the Nematostella genome using the CLC Bio genomics workbench version 5.1 NGS tool mapper. There is a sam file and an indexed sam file. When placed in the same folder, they can be viewed as tracks on top of the Nematostella genome in a viewer such as the Broad Insitute's Integrative Genomics Viewer: (http://www.broadinstitute.org/igv/).
